Funder"},"subitem_funder_names":[{"subitem_funder_name":"日本学術振興会","subitem_funder_name_language":"ja"}]}]},"item_19_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1996-03","bibliographicIssueDateType":"Issued"}}]},"item_19_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"最近、がんの発生におけるフリーラジカルの役割については多くの注目を集めてきているが科学的な根拠については未知の点も多い。特に、防御機構が正常に機能しない状態に陥ったときに生体内では過剰な活性酸素が生成し、フリーラジカル連鎖反応が誘発される。その結果、生体膜での傷害や生体重要物質での損傷にもとずく機能傷害の蓄積と共に遺伝子レベルにおける傷害が「遺伝毒性」を発現し、「がんの発生」を誘発したり「がん化の促進」に大きな役割をはたしているのではないかと考えられてきている。このような遺伝子レベルにおける酸化的傷害を抑制すること、特に食品成分が抑制因子となりうるかについてを評価するために、老化に関連した疾病のマーカーとして酸化修飾塩基の代表である8-デオキシグアノシンのモノクローナル抗体による検出法の応用を日本老化制御研究所 (越智)との共同研究で開発することができた。そこで本年度は、まず、ゴマ種子中に大量に含まれている新規な抗酸化前駆体であるリグナン配糖体の持つ酸化傷害予防効果の検討を行った。方法は、ゴマ脱脂粕をビタミンE欠乏飼料中に10%加えてラットに2ケ月間投与し、四塩化炭素で過酸化傷害を与えた。その結果、ビタミンE欠乏食だけの場合に較べてゴマ脱脂粕の場合は肝臓や血液中の過酸化度と共に尿中での過酸化度、さらに8-デオキシグアノシン量を有意に低下させた。これらの結果より、セサミノール配糖体が腸内細菌により加水分解を受けた後に生体内に吸収され抗酸化的な防御効果を示すという新しい機構を提出することができた。また、本年は、生体膜の酸化的傷害物質として代表的な4-ヒドロキシノネナールのモノクロナール抗体の作製に成功し、さらに最近マロンジアルデヒドにより修飾されたタンパク質に対するポリクローナル抗体の作製にも成功している。これらの抗体による組織染色、特に酸化傷害により生じた腎臓がん部位での検出への応用にも成功している。さらに、培養細胞系、特に、チャイニーズハムスターや大腸由来の細胞で過酸化水素を中心とした酸化的傷害の保護効果についても、新しい機構を提出することができた。また、ゴマ油中の抗酸化成分として抗酸化的な防御効果の検討を行ったセサミノールについては、ビタミンEに対して強い相乗的な効果が見られるという新しいメカニズムを明らかにすることができたので、これらのリグナンを食品因子として代謝も含めた新しい研究へのアプロ-チが期待される。","subitem_description_language":"ja","subitem_description_type":"Abstract"}]},"item_19_description_5":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"科学研究費補助金
