Funder"},"subitem_funder_names":[{"subitem_funder_name":"日本学術振興会","subitem_funder_name_language":"ja"}]}]},"item_19_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2001-03","bibliographicIssueDateType":"Issued"}}]},"item_19_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"本研究は、生活習慣病、なかでも、若年層も含めて大きな問題となりつつ糖尿病の合併症で最近特に注目を集めている酸化ストレスの影響について分子レベルでの解明と防御を目的として、まず、酸化ストレス評価法として好中球の過剰発現由来のジチロシンに特異的なモノクローナル抗体、脂質ヒドロペルオキシドによる酸化修飾タンパクやDNAに特異的なモノクローナル抗体の作製に成功した。さらに、明らかな高血糖を呈する2型糖尿病モデルであるAkita mouseの酸化ストレス状態を解析することを目的とし、血液および組織の脂質過酸化やタンパク質の酸化障害を検討した。その結果、脂質過酸化初期反応生成物であるリノール酸13-ヒドロペルオキシドとリジンとの付加体であるNε-(hexanonyl)lysine(HEL)、および、タンパク質の酸化障害のバイオマーカーであるdityrosine(DT)が顕著に増加していることを明らかにした。これらの結果は、高血糖状態が生体内での酸化ストレスを亢進していることを示唆するもので、糖尿病合併症予防の可能性について検討を行う目的で、カレー料理に一般的な香辛料、ターメリックの黄色色素、クルクミンとその生体内代謝物であるテトラヒドロクルクミンの糖尿病合併症予防効果について検討を行った。その結果、オスのSDラット(n=4)に25%ガラクトースを含んだ飼料を与え、同時にクルクミン、テトラヒドロクルクミンは0.2%餌にまぜて摂取させたところ、6週間後に生じた末期的な白内障は顕著に抑制された。そこで、テトラヒドロクルクミンに焦点をあててサルを用いたin
