1.datAの機能へのIHFの関与 datA内に存在するIHF認識配列にIHFが非常に高い親和性を示すことが判明した。DnaA boxとIHF結合部位、およびそれらの近傍にいろいろな変異を導入した株の解析から、IHFにより折り曲げられたdatA領域全体に、秩序正しい3次元構造の変化が引き起こされ、多量のDnaA分子が結合するというモデルが示唆された。 2.dnaA遺伝子の染色体上の位置と複製開始制御の関係 dnaA遺伝子は染色体上でoriC近傍に位置し、oriCとともに複製開始後しばらく細胞膜に隔離される。この間は、転写が抑制されている。dnaA遺伝子の位置を本来の位置から移動させると、細胞膜への隔離の時期がずれ、細胞周期における転写抑制の時期が本来の時期とずれることになる。このような株を系統的に作成、解析した。その結果、dnaA遺伝子の隔離が複製開始制御に関与することが示唆された。この結果はこれまでの一般的な考えとは異なるものである。 3.DnaA-GFP融合タンパク質に依存して生育する株の構築細胞周期における細胞内のDnaAの挙動を知る目的で、染色体上のdnaA遺伝子内のドメインIIの一部をGFPに置換した株を作成した。この株は DnaA-GFP融合タンパク質に依存して生育した。現在、蛍光顕微鏡によりDnaAの挙動を詳細に解析中である。","subitem_description_language":"ja","subitem_description_type":"Abstract"}]},"item_19_description_5":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"科学研究費補助金
