To see the boundary between the blastodisc and the yolk vacuole more clearly, we microinjected FITC-dextran into the yolk vacuole. To monitor mitotic events, we either microinjected Hoechst 33342 into living eggs or stained fixed eggs with Hoechst 33258; these eggs were monitored via a SIT camera and video monitor. We found that the first sign of cytokinesis is the formation of an internal furrow that forms at the surface between the blastodisc and the yolk vacuole.
