Bone collagen is commonly used for 14C-dating of bones. The bone pretreatment includes the removal of mineral phase of bones with 0.4-0.6M HCl followed by a gentle-base-acid extraction to remove extraneous carbon such as humic acids from the organic bone residue. Next, the collagen is dissolved as gelatin at 85-90℃ and pH 3 and, and filtered through a glass filter to remove non-soluble contaminants, then freeze-dried. The gelatin with its yield of <0.7 wt% could remain exogenous carbon, and so more cleaning step using the high molecular weight (>30 kD) of gelatins to remove contaminants by ultrafiltration is needed. The appropriate schematic overview of collagen extraction and ultrafiltration is performed., 遺跡から出土する骨は、当時の人の食性や生活環境、年代などの情報を直接与えてくれるものとして、考古学において非常に重要である。骨の14C年代測定においては、一般的に、硬タンパク質の骨コラーゲンが用いられる。骨は、まず0.4-0.6M HClによって、無機成分を除去する。得られた酸不溶脱灰成分に0.1MNaOH、0.1MHClにより穏やかにアルカリ・酸処理を行ない、フミン酸などの外来炭素を除去する。次に、85-90℃、pH3の弱酸性の水にコラーゲンを溶解し、ゼラチン抽出を行なう。ゼラチン溶液はガラスフィルターによるろ過を行ない、不溶性の外来炭素を除去する。得られたゼラチン収率が0.7wt%よりも低い場合は、外来炭素が完全に除去されていない可能性が考えられ、さらに限外ろ過によって、3万分子量以上の高分子ゼラチンのみをふるい分けする必要がある。本稿においては、以上の骨コラーゲン抽出法、およびゼラチンの限外ろ過法に関して、現在最も適切と考えられる手順について紹介する。}, pages = {185--189}, title = {14C年代測定のための骨試料調整法}, volume = {23}, year = {2012} }
