The effect of decline in water potential of the root medium (0 to -14 bars) on water and mineral (N, P, K, Ca, Mg, Na) uptake, transpiration, and growth was investigated for Pinus thunbergii yearlings about 100 mg each in fresh weight. Polyethylene glycols – 400 was used as the osmotic substrate. Water uptake, transpiration and growth were decreased as water potential of the root medium was reduced. The growth of pine yearlings at -10 bars was stopped and at -14 bars half of yearlings were dead during 14 days. The rate of water uptake -10 bars was reduced to 13% of the rate at 0 bars. The transpiration rate at -10 bars was decreased to 62% of the rate at 0 bars. The time course of the water uptake at each levels of water potential followed approximately the Mitscherlich curve, whose parameters, the maximum water uptake (A) and the coefficient of water uptake (λ) were correlated closely to the water potential of the root medium. Decline in water potential of Pine root medium (-0.6, to -6.5 bars) reduced P uptake. Absorption of other minerals (N, K, Ca, Mg, Na) did not show significant change. Diurnal patterns in leaf water potential of water deficit were examined with a pine tree in summer clear days. Variations in water potential and water deficit during the day were likely related to changes in evaporative demand of the air. Water deficit-water potential relationships were not same for current year leaves and one year old leaves. In one year old leaves, the relationships shifted to lower potential for given water deficit. 2. Salinization (NaCl, KCl, MgCl_2, MgSO_4, -3.0 to -6.5 bars) reduced growth of pine yearlings. Especially Mg reduced growth of pine yearlings and added NaCl to the medium reduced Ca uptake. 3. The effect of water stress on the structure of organells of pine leaf cells was analysed for the juvenile leaves floating on polyethylene glycols solution by an electron-microscopy. Dehydration (-8 bars, 4 days) induced changes in submicroscopic structure of the cells. The most pronounced changes were chloroplast fusions and increase in number of fine vesicles. They seemed to take place during the plasmolysis and deplasmolysis, which were respectively induced by the dehydration and by washing leaf material with water. Severe dehydration (-15 bars, 4 days) destroyed almost completely all organells of the pine leaf cells and as a result the cells seemed to lose their ability to be deplasmolysed. 4. Water potential of the rhyzosphere (0 to 5 cm depth) of pine yearlings falled to -18 bars in summer clear days. Disappearance of pine yearlings on sand dunes seems to be caused by drought of summer. 5. From these results the problems of planting under dry and saline conditions were discussed.
